How did researchers find out the effects of Excitotoxins?

“Excitotoxins cause a brain cell to become very excited and your neurons basically fire spastically until they finally burn out,” – Dr Lippman.
My question is, how did they practically come to that conclusion, I scoured the internet they never say how the experiments were made.

the wiki article was helpful. but I’m not sure like how the damage is done, when the blood reaches the brain or when the tongue tastes the food.
