How can this be proven to kill us?

Dr Mercola, an osteopath, has an article I was reading that stated if you sit for eight hours a day you’ll die much earlier than if you stand up as much as possible. He also said each hour you should get up and walk around and that no exercise that you do will cancel out this danger of sitting all day.
How can this have been proven?
